Actions:In vitro,The
IC50 of total alkaloids and tylocrebrine on S180 cell line
were 0.1~1.0 ug/ml and tylophoridine was 0.05~0.1uug/ml.
In vivo, the inhibiting rates of
total alkaloids at 4mg/kg*8,im. and ip. against S180 sarcoma
were 66.9% and 56.3%(P<0.01); tylophorindine at 7.0mg/kg*8,ip.
and tylophorindine at 2.0mg/kg*8,ip their inhibiting rates
against S180 were 33.0% and 54.2%(p<0.01) respectively.
The inhibiting rates of total alkaloids at dose of 4mg/kg*8
and 4.5mg/kg, ip. against U14 were 31.2% and 45.8%. The life
span prolongating rate of total alkaloids at dose of 7mg/kg¡Á6,
ip. against L615 was 33.3%. The inhibiting rates of total
alkaloids at 2.3mg/kg*8, ip. against W256 was 64%. The life
span prolongating rates of total alkaloids against L1210 and
P388 were 51% and 70% respectively.
Indications:It
could be used for treatment of leukemias and lymphosarcoma.
118 cases of various kinds of leukemias have been treated
with total alkaloids prepartion, among the evaluable 52 cases,
the single drug treatment group has 16 cases, in which PR
7,the effective rate was 43.7%;the combined group(with prednisone)
has 31 cases, in which CR 4 (acute lymphocytic leukemia),
PR 17, the total effective rate was 67.7%. The results showed
that its therapeutic effect was better for children leukemias
than that of adult leukemias. The combined therapy with prednisone
and successive drug treatment were better than of intermittant
or single drug treatment.
The dose of the drug: for adult, 10~20mg
a day, for children 0.2~0.4mg/kg, after diluted with 5% glucose
solution, administered by iv-drip administration.
The adverse reactions of the drug commonly
were digestive tract side effects, such as anorexia, nausea,
vomiting and diarrhea.[20:121].
