Actions:Studies
in vitro indicated that As2O3 had double effects on the acute
promgolocytic leakemia (APL) cells inducing apoptosis at higher
drug coucentrations (0.5~2.0umol/L) and inducing partial differentiation
at lower drug concentrations (0.1~0.5umol/L). As2O3. (0.1~2.0umol/L)
could rapidly modulate and degrade APL-specific marker molecular
PML-RARa protein. As2O3. might interfere the signal transducton
of BCR/ABL protein by reducing its PTK activity of cytosol
and membiane proteins abd BCR/A- BL protein the tyrosine phisphorglation
proteins declined and induced K56- 2 cells apoptosis. In addition,
As2O3. had a remarkable cytotoxic effect on SGC-7 9.1. The
cytotoxic rate of 10umol/L As2O3 on SGC-7901 exhibited morplologic
features of apoptosis. The apoptotic index was 7%~15.1% by
TUNEL and FACS assay. As2O3. could inhibit significantly proliferation
of gastric cancer cell SGC-7901 and induce apoptosis, so it
had a potential valne in the treatment of gastric cancer.
Indications:The
Gan-Ling No. I injection solution (1 ml containing As2O31mg)
was made by the first affiliaed hospital of Ha-Er-Bin medical
university , direction: Gan -Ling No. I 10ml was added into
500ml 10% glucose solution for intravenous drip, once a day.
26 days as a course of treatment, interval 7~14 days made
the next course of treatment .The drug was used for the treatment
of 32 cases of acute promyelocytic leukemia. The results showed
that 21 cases were completely remission (65.6%), 4 cases were
partial remission (12.5%), the total remission rate was 78.6%
. As2O3 was prepared in arsenous acid injection and used for
the treatment on 224 cases of acute promyelocytic lenkemia.
The results showed that the remission rate was 91%, the survival
rate of five years was 54%, 13 cases have been survived over
10 years, the most longest life span reached 26 years. In
addition this preparation also had significanfly effects on
pancreatic cancer, gastric cancer, hepatoma, megakaryocytic
lenkemia and B-lymphocytoma.
